糖尿病越漸流行,也是台灣十大死因之一。麵條作為亞洲人的主食,其主成分以碳水化合物為主,使麵條具有高升糖指數(Glycemic index;GI),長期食用會增加二型糖尿病發生風險,因此開發低GI 麵條越受重視。紅茶及薑科植物(生薑、月桃)在許多研究中顯示具有抗氧化、抗發炎、抗癌等功效,使其越受關注。本研究採用大吉嶺紅茶葉、生薑與月桃葉為原料,分別以水及 50%乙醇萃取,利用清除(1,1-二苯-2-三硝苯肼;DPPH)、(2,2'-azinobis-3-ethyl benzothiazoline-6-sulphonic acid;ABTS)自由基能力和還原力(Ferric reducing a

ntioxidant power;FRAP)試驗來評估萃取物之抗氧化能力;再藉由抑制 BSA-glucose 糖化反應終產物(Advanced Glycation End Products; AGEs)試驗,及抑制α-glucosidase和α-amylase活性試驗評估降血糖潛力;最後分析其機能性成分,篩選出最具潛力之萃取物並探討其添加於麵條後之物性分析、GI值表現與消費者感官喜好。結果顯示,抗氧化能力DPPH以紅茶50%乙醇萃取物(BT-50)之IC50值:11.24μg/mL及FRAP以紅茶水萃取物(BTH)之EC50值:49.61 μg/mL為最佳;降血糖潛力AGEs以BTH之IC50

值:296.11 μg/mL及α-glucosidase以BT-50之IC50值:3.33 μg/mL為最佳,綜觀上述其抗氧化及降血糖潛力均以紅茶葉>月桃葉>生薑葉顯示。測定總多酚類後發現,總多酚(TPC)含量以月桃葉50%乙醇萃取物(AZL-50)為最高(約為80 mg/g);總黃酮類(TFC)則以生薑葉50%乙醇萃取物(ZOL-50)為最高(約為54 mg/g),表示生薑與月桃葉也具有良好之應用潛力。而在多酚類組成分分析、總三萜類及茶黃素分析上分別以BTH (4.97 mg/g)、BT-50 (111.41 mg/g)與BT-50 (15.96 mg/g)最高,皆以紅茶葉表現最佳,因此以紅

茶作為麵條食品之添加原料。在物性分析、體外澱粉消化試驗結果顯示,添加 3%紅茶葉粉(BTP-3)能夠在較低影響麵條的質地下賦予其具有最佳之體外澱粉消化表現。感官品評試驗中使用消費者型品評員來評估七種不同配方紅茶麵條(分別添加紅茶葉粉0、1、3、5、7 %以及使用紅茶茶湯取代水再額外添加0、3 %茶粉)的感官描述特性,以及了解消費者之間的喜好差異。研究結果顯示,消費者整體接受度隨著紅茶葉粉添加比例的提高而略微下降,僅7%紅茶麵條(BTP-7)以外其餘皆與無添加之生麵條(BTP-0)無顯著性差異,表示麵條作為紅茶健康食品開發之載體是可以被國人飲食文化所接受。在描述性分析中發現影響消費者對紅茶麵條具

有正面影響之特性包含順口感、茶味(剛剛好)、具有彈性、健康的、天然的及口感純粹等,而綜觀各項分析結果,以添加3% (BTP-3)之紅茶麵條具有最符合消費者所期望之特性,同時BTP-3其硬度、內聚性、彈性與咀嚼性皆與生麵條無顯著性差異,因此,於麵條中添加3%之紅茶葉粉能夠賦予麵條具有最佳之低GI值表現,同時也最具有符合消費者期望的感官特徵。

Gut bacterial β-D-glucuronidases (GUSs) catalyze the removal of glucuronic acid from liver-produced β-D-glucuronides. These reactions can have deleterious consequences when they reverse xenobiotic metabolism. The human gut contains hundreds of GUSs of variable sequences and structures. To understan

d how any particular bacterial GUS(s) contributes to global GUS activity and affects human health, the substrate preference(s) of individual enzymes must be known. Herein, we report that representative GUSs vary in their ability to produce various xenobiotics from their respective glucuronides. In a

n attempt to explain the distinct substrate preference, we solved the structure of a bacterial GUS complexed with coumarin-3-D-glucuronide. Comparisons of this structure with other GUS structures identified differences in TIM barrel loop 3 (or the 2-helix loop) and loop 5 at the aglycone-binding sit

e, where differences in their conformations, hydrophobicities and flexibilities appear to underlie the distinct substrate preference(s) of the GUSs. Additional sequence, structural and functional analysis indicated that several groups of functionally related gut bacterial GUSs exist. Our results pin

point opportunistic gut bacterial GUSs as those that cause xenobiotic-induced toxicity. We propose a structure-activity relationship that should allow both the prediction of the functional roles of GUSs and the design of selective inhibitors. Furthermore, we demonstrated how charge, conformation, an

d substituents of several uronic iminocyclitols and analogues contribute to the inhibitory potency and selectivity for gut bacterial GUSs by using crystallographic and biochemical methods. Uronic isofagomine was a potent inhibitor (Ki up to 4 nM) unlike uronic deoxynojirimycin that showed a less pot

ent inhibition (Ki up to 930 nM), indicating that the positive charge at the anomeric position, but not endocyclic oxygen position, was favorable to form electrostatic interactions with the two catalytic glutamates of GUSs. Moreover, D-glucaro-δ-lactam displayed good inhibitions (up to 280 nM) for b

acterial GUSs owing to the half-chair conformation that favored several polar contacts and a charge-dipole interaction. Additionally, C6-alkylated uronic isofagomine displayed selective inhibition for opportunistic bacterial GUSs, which was attributed to the hydrophobic interactions between the prop

yl group and the loop 5 residues of the GUSs.In contrast, N1-alkylated analogue was a non-selective and moderate inhibitor for GUSs, which was explained by a steric hindrance between the alkyl group of the inhibitor and the catalytic acid/base glutamate of GUSs. In summary, we identified the respons

ible group of GUSs for the xenobiotic-induced toxicity, and developed a selective inhibitor targeting these GUSs.
